Optimizing the Conditions of Inducing Polyploid of Fritillaria cirrhosa and Content Determination

The induction rate of optimized conditions of Fritillaria cirrhosa including the concentration of colchcine,the time of being soaked in colchcine,the volume fraction of dimethyl sulfoxide and temperature was determined by orthogonal text. A optimal combination was screened through the crossed test of L9(34). The order of factors affecting the induction rate was the concentration of colchcine the time of being soaked in colchcine the volume fraction of dimethyl sulfoxide temperature.The optimal combination was the concentration of colchcine 900 mg / L,the time of being soaked in colchcine 48 h,the volume fraction of dimethyl sulfoxide 4%,temperature 20 ℃, with induction rate of 94.3%. Compared with the chromosomal number,it was proved that the callus of F. cirrhosa was polyploidy. The contents of total bio-medicinal ingredients with wild,tissue culture of diploid and polyploid bulbs of F. cirrhosa were effectively determinated. The total alkaloid content of polyploid bulbs of F. cirrhosa was the highest with up to 0.657%.
