Objective:To study the effects of culture components and illumination on callus growth and synthesis of flavonoids of Astragalus membranaceus(Fisch.) Bge.Methods:Adjusted the concentration of carbon, nitrogen, phosphate and calcium in the culture solution, and kept the astragalus callus under dark or illumination, then detected the content of flavonoids by spectrophotometry. Results:The optimum condition of total flavonoids of astragalus accumulation was(24±1)℃ in the dark and the basic medium was Murashige and Skoog(MS)(plus 0.5 mg·L-12,4-D and 1.0 mg·L-16-BA) with 80 g·L-1sucrose,1.5 mmol·L-1KH2PO4 and 1.496 mmol·L-1CaCl2,the proportion of NH+ and NO3-was 2∶1(nitrogen concentration is 60 mmol·L-1).Conclusion:Sucrose,selected as source of carbon is more favored for the callus growth and the synthesis of saponins than glucose and maltose are.And mixed nitrate is favored for the synthesis of flavonoids compounds.
