OBJECTIVE To establish a simple and accurate HPLC method for the study of lactone accumulation of A.paniculata and to study the correlations of the lactones with other characters.METHODS Detected at 226nm,analytes were separated on a Yilete ODS C18 column(150 mm×4.6 mm,5 μm) eluted gradually with methanol and water(18:82),with methanol gradually increased from 18% to 60% in the whole analysis.After determination,SPSS 13.0 software was used to calculate the correlations of lactones with other characters.RESULTS Under the above conditions,the lactone HPLC method of Andrographis paniculata was constructed,and through the software SPSS 13.0,the correlations of andrographolide and 14-deoxy-11,12-dehydroandrographolide with other characters were calculated.Through determination and analysis,it was found that during the growth period the contents of andrographolide varied greatly,it accumulated rapidly in the early growth period,and reached highest level at initial flower development period(about 150th day of its growth);while the accumulation of 14-deoxy-11,12-dehydroandrographolide was very slow.The analysis of correlation coefficients suggested that the contents of andrographolide were significantly correlated with leaf number,dry weight,plant height,dry weight of leaf istics,alcohol extract and growth time;while the content of 14-deoxy-11,12-dehydroandrographolide had almost no correlation with these characters.CONCLUSION The developed method for analysis of the lactones,accurate and reproducible,was suitable for the quality evaluation of Herba Andrographitis.
