[ Objective ] To explore the effect of on androgen binding protein( ABP )gene expression in rat Sertoli cells. [ Methods ] The survival rate of Sertoli cells of primary culture for 4-5 days was counted after incubated with DMSO, 30, 50, 80, 100 umol/L for 24h respectively. And the androgen binding protein gene mRNA expression levels in Sertoli cells incubated with DMSO and of 10, 30, 50 μmol/L for 24 h respectively were detected by one-step RT-PCR method. [ Results ] The survival rate of Sertoli cells of primary culture was above 90% in those groups of 50 μmol/L and lower, whereas in higher dosage groups Sertoli cells survival rate decreased sharply. The gray density ratios of androgen binding protein gene mRNA in the groups of DMSO, 10, 30, 50 μmol/L were 0.314 0±0.102 0, 0.392 0±0.094 9, 0.583 710.122 1 and 0.649 0 10.171 8, respectively, and the ratios were ascendant with increase of concentration in Sertoli cell cultures, and there was a significant dose-effect relationship. [ Conclusion ] might enhance the expression of ABP gene, which might he a protective adaptation attributing to the lower androgen in Sertoli cells because of p,p'-DDE s competitive binding androgen receptor with androgen.
