Objective: To detect the relationship between the presence of treponema denticola(Td) and chronic periodontitis. Methods: Subgingival plaque samples were taken from disease sites and healthy sites of 58 patients with chronic periodontitis and treated with 1%Triton X-100 and 100℃ for 10 minutes to prepare DNA templates. The fragments of 16s rRNA gene detected by PCR. Result: 47 of 58 (81.0%) subgingival samples from disease sites and 8 of 58 (13.8%) healthy sites showed positive PCR results. Conclusion: (1)The detection rate of Td in disease sites was much higher than the healthy sites, it must be the putative etiologic agent in chronic periodontitis and Td infection was close related to the severity of chronic periodontitis.(2) PCR was sensitive and distinctive method to detect Td.
