Objective To explore the optimal extraction process and determination method of magnolol and honokiol in leaves of Magnolia officinalis Rehd.et Wils in Fujian.Methods The orthogonal design was used to optimize the extraction process.The influential factors of extraction rate were analyzed.The contents of magnolol and honokiol were taken as the index for investigation and determined by HPLC.Results The highest extraction rate could be obtained with the ultrasonic extraction method,and the optimal conditions for the extraction by using methanol as solvent were obtained as follows the ratio of material to solvent 1:75 at 50℃ for 20 min.The linear range of magnolol was 10-60 μg.mL-1(r=0.9996).The linear range of honokiol was 20-120 μg.mL-1(r=0.9997).The content of magnolol was 0.09%,honokiol was 0.16% and the average content of total phenol was 0.25%.Conclusion The method is simple,stable and accurate,and suitable for the extraction and determination of magnolol and honokiol in leaves of Magnolia officinalis Rehd.et Wils.
