This experiment was conducted to study the effect of different detective methods on the concentrations of airborne microbes.Following air samples were collected by AGI-30 air sampler,this study was to compare the concentrations of airborne microbes in the livestock and poultry houses by using the cultivation and counting cells after DAPI-staining(4,6-diamidino-2-DAPI) directly,respectively.The concentrations in poultry,pig and cow houses were within a range of 5.73×105~6.72×106 cfu/m3 air,9.5×105~4.01×106 cfu/m3 air,5.4×104~8.33×105 cfu/m3 air,respectively.The direct counting concentrations were between 8.0×106~3.25×108 cell/m3 air,1.5×107~2.28×108 cell/m3 air,9.0×105~5.93×107 cell/m3 air,respectively.The concentrations of direct microscopic counting cells after DAPI-staining were 9.6 to 2500 times higher than those of counting colonies.DAPI-staining counting method could reflect the accurate concentrations of airborne microbes in the environment,and assess healthy risks scientifically.
