Objective
To observe the expression of CD69 molecules in γδΤ cells activated by human peripheral blood phorbol myristate acetate(PMA) and ionomycin(IM).
Methods
Healthy human PBMC was isolated.PMA and IM were used to stimulate PBMC.We used flow cytometry to detect the dynamic expression of CD69 in CD3+ T cells and γδΤ cells after0, 6, 12, 24, 48 and 72 hours.
Results
PBMC was stimulated by PMA and IM for 0, 6, 12, 24, 48 and 72 hours.After that, CD69 expression in CD3+ T cells was 1.0%, 99.5%, 99.1%, 98.0%, 93.3% and 92.1%, respectively.CD69 expression in γδΤ cells was 0.91%, 99.3%, 98.6%, 93.6%, 88.7% and 87.0%, respectively.
Conclusion
After being stimulated by PMA and IM, CD3+ T cells and γδΤ cells were activated rapidly.The expression of CD69 was close to 100%, and the highest expression was found at 6 hours.After 6 hours, the CD69 expression fell down slowly.The similarly improve effect of CD69 expression was observed in both CD3+ T cells and γδΤ cells.
