Background: The immuno-pathogenetic mechanism of Helicobacter pylori (H.pylori) on gastric mucosa is much complicated. The effects of different protein molecules of H.pylori on the human body are urgently needed to know. Currently the studies on the outer membrane of H.pylori are very few. Aims: To set up the two-dimensional gel electrophoresis (2-DE) and computer-assisted image analysis for the proteomics study of H.pylori outer membrane proteins, and to compare the difference of the 2-DE images between the outer membrane proteins of H.pylori stains related to patients with different gastric diseases, and to provide the technical basis of proteomics research of H.pylori outer membrane proteins. Methods: To select and separate the two H.pylori strains (H.pylori 185 and H.pylori 161) from samples of patients with different gastric diseases, and separate the outer membrane proteins of the two H.pylori strains. The H.pylori outer membrane proteins were extracted and analyzed by computer-imaging analysis software. Results: By duplicating the 2-DE and imaging analysis of the H.pylori outer membrane proteins, the matching rate of protein spots were 98%, most of proteins were superposition well, the repeat rate and resolving power were higher. Among the 116 (main protein spots were 39) and 129 (main protein spots were 48) protein spots in H.pylori 185 and H.pylori 161, 35 protein spots of them were found to be the same. Conclusions:The outer membrane proteins of H.pylori strains of different gastric diseases have many protein spots in common and also have their own specific protein spots, with slight discrepancy in the protein content. We may conclude that the 2-DE is a useful method to study the proteomics of H.pylori outer membrane proteins. Analysis of outer membrane protein of the H.pylori strains probably may provide of the theoretical basis for the diagnosis and treatment of H.pylori-associated diseases.
