Objective To understand the changes in biological characteristics of hPDLPs under induction of EMD.Methods Cultured human periodontal ligament cell populations(hPDLPs) were exposed to the conditioned culture media containing 100 mg/L Emdogain(EMD) for six days.Cell morphology was observed under light microscope, and cell morphology in the cementum chip was observed by in scanning electron microscope before and after induction.Expression of bone sialoprotein, type Ⅰ collagen protein was detected immunohistochemically. Results After induction, cell morphology of hPDLPs changed from a long fusiform to polygon, like cementoblasts rather than the fibroblasts.Immunohistochemistry showed that expression of bone sialoprotein, type Ⅰ collagen was significantly increased. Conclusion EMD could induce differentiation of hPDLPs to cementoblasts. It provide the basis for further research into the regeneration of periodontal tissues and cementum generation.
