EFFECT OF QUERCETIN ON FOAM CELL FORMATION AND THE EXPRESSION OF ATP BINDING CASSETTE TRANSPORTER AI

Objective To observe the effect of quercetin on foam cells formation and the expression of ABCAl(ATP binding cassette transporter Al).Methods RAW264.7 cells were incubated with l00mg/L oxidzed-low density lipoprotein(ox-LDL) for 48h to establish foam cell model,then the effects of different concentration of the quercetin on the formation of macrophage-derived foam cells were observed.The cellular lipid accumulation was examined by oil red O staining.ABCAl expression was evaluated by immunofluorescence and Western blotting.Results Oil red O staining showed that intracellular lipid droplets in model group were significantly more than those in control group,while those in quercetin groups were fewer than those in model group.The green fluorescent intensity in model group was significantly higher than that in control group,and increased significantly after interference of quercetin with different concentrations.The protein expressions of ABCAl in model group were significantly higher than those in control group(P0.0l),but increased significantly after interference of quercetin with different concentrations(P0.0l) dose-dependantly.Conclusion Quercetin inhibits atherogenesis and suppresses foam cell formation in macrophage via ABCA1 upregulation.
