Pathway Analysis for Production of L-Histidine by Corynebacterium glutamicum TL1105

Objective: Pathway analysis was conducted to determine theoretically optimal mode for the L-histidine production by Corynebacterium glutamicum TL1105.Methods: METATOOL was used to analyze the L-histidine biosynthesis pathway in C.glutamicum TL1105.Results: Theoretically optimal mode and theoretical L-histidine biosynthesis yield of 1.2 were determined.The phosphoribosyl pyrophosphate(PRPP) was found to be the key node of L-histidine biosynthesis pathway by analyzing all the different elementary modes and the flux of L-histidine could be largely increased by changing the environmental factors.The production of L-histidine in fermentation broth increased 39.2% with the addition of L-glutamate than that in the original conditions.Conclusion: According to the pathway analysis result,the production of L-histidine can be largely increased by changing the environmental factors.
