Effects of Serum Containing Gukang on Osteoprotegerin and Receptor Activator of Nuclear Factor-κB Ligand in the Cell Co-culture System

Objective:To study the effect of serum containing Gukang on osteoprotegerin(OPG)and Receptor Activator of Nuclear Factor-κB Ligand(RANKL)in the cell co-culture system.Methods:We observed changes of OPG,RANKL and their ratio in osteoblast-osteoclast culture supernatant.Results:After adding serum-free medium,since the 3rd day,OPG content in Gukang group was significantly higher than that in the blank group(P0.01),RANKL content in Gukang group was significantly lower than that in the blank group on the 7th day(P0.05),but OPG and RANKL in the blank group had no significant change in the culture period.Since the 3rd day,the ratio of OPG/RANKL in Gukang group was significantly higher than that in the blank group(P0.05,P0.01),the ratio of OPG/RANKL on the 5th and 7th day was higher than that on the 1st day in Gukang group(P0.05),but the ratio of OPG/RANKL in the blank group had no significant change in the culture period.Conclusion:Serum containing Gukang is effective on osteoblast and osteoclast,it can regulate the ratio of OPG/RANKL in cell survival environment,and indirectly affect the functional activity of osteoclast.
