This experiment used isoelectric focusing electrophoresis(IEF)and HPLC methods, through the determination of the rhIGF-1 isoelectric point as an example, established a rapid determination of protein isoelectric point method. The gel was 7.5% of the gel concentration, 3% of the crosslinking degree, and 6% of the Amoholine, and determined the isoelectric point of rhIGF-1 by the Alpha imager TM 2200 and HPLC. The results showed that the isoelectric point of rhIGF-1 was 8.20.The pH gradient curve linear regression equation was Y=0.0664X+3.8217, The correlation coefficient r was 0.9956. That was mean the result was accurate.
