Effects of low oxygen on expression of mRNA of HIF-1α,SDF-1 and VEGF in hBMSCs

Objective To observe the effects of low oxygen on the expres sion of mRNA of hypoxia-inducible factor-1α(HIF-1α),stromal cell derived factor-1(SDF-1) and vascular endothelial growth factor(VEGF)in hBMSCs.MethodThe third passages of hBMSCs were cultured in 1%O2 and 20%O2, HIF-1α,SDF-1 and VEGF mRNA were detected by RT-PCR. ResultsCultured under 1%O2 condition, the mRNA of HIF-1α,SDF-1 and VEGF of hMSCs increased greatly(P0.01).There was obvious association between the expression of HIF-1α,SDF-1 and VEGF mRNA(P0.01),but no correlation were found between SDF-1 and VEGF mRNA expression.ConclusionLow oxygen induces the expression of HIF-1α,and results in increasing SDF-1 and VEGF expression in hBMSCs.
