A Modified Plate Transformation Method of Escherichia coli by Plasmids

A modified plate transformation method of Escherichia coli by plasmid was described.Recipient Escherichia coli cells were stored at -30 ℃ in PEG,MgSO_4 and glycol solution,then plasmids with different molecular weights were mixed with these cells,and transformed and selected directly in CaCl_(2) plates.It was showed that the modified plate transformation method was of higher transformation efficiency and maintenance than classic CaCl_2 method when transformed by plasmid of higher molecular weight.
