Simultaneous determination of caffeic acid and chlorogenic acid in Pugongying Ruangao by HPLC

Objective To establish a method to determine caffeic acid and chlorogenic acid in Pugongying Ruangao. Methods HPLC method was used with Luna C18(250 mm×4.6 mm,5 μm)column and the mobile phase consisted of acetonitrile-0.02 mol·L-1 NaH2PO4(14∶86).The flow rate was 1.0 mL·min-1,the column temperature was 30℃,and the UV detector wavelength was 323 nm. Results The linear ranges of caffeic acid and chlorogenic acid were 0.81-16.20 μg·mL-1(r=0.999 8)and 3.22-64.30 μg·mL-1(r=0.999 6),respectively.The average recovery of caffeic acid and chlorogenic acid was 98.8% and 98.0%,respectively. Conclusion The method is simple,repeatable and accurate,which can be applied in the quantitative determination of caffeic acid and chlorogenic acid in Pugongying Ruangao.
