Objective To study the expression of Ang-1 /Tie-2 in peripheral blood endothelial progenitor cells( EPC) in patients with hypertension,and the its role in the regulation of number and function of EPC. Methods Total 30 patients with essential hypertension( observation group) and 30 health people( control group) in our hospital in 2012 were enrolled in the study. The amount and proliferation activity of peripheral blood endothelial progenitor cells were measured,the levels of Ang-1 /Tie-2 were tested by RT-PCR and Western-blot simultaneously. Results The amount and proliferation activity of peripheral blood endothelial progenitor cells in the observation group was( 6. 17 ± 0. 89) × 103/L and( 0. 78 ±0. 25),lower than( 6. 94 ± 1. 12) × 103/L and 1. 14 ± 0. 36 in the control group( P 0. 05). The colorimetric optical densities( OD) of Ang-1 and Tie-2 were 1. 65 ± 0. 97 and 4. 32 ± 0. 84 in the observation group,was lower than 2. 37 ±1. 52 and 5. 52 ± 1. 18 in the control group( P 0. 05). The levels of Ang-1 and Tie-2 protein in endothelial progenitor cells were 0. 06 ± 0. 02 and 0. 09 ± 0. 04 in the observation group,lower than 0. 21 ± 0. 05 and 0. 20 ± 0. 03 in the control group( P 0. 05). Conclusion The amount and activity of endothelial progenitor cells decreased,while the expression of Ang-1 /Tie-2 also down-regulated in patients with hypertension,this promotes that the down-regulation of Ang-1 /Tie-2 may probably lead to the changes of endothelial progenitor cells.
