Objective:To study the effect of Hirudin on contractibility of hypertrophic scar model,fibroblasts' proliferation and secretion in vitro.Methods:Human hypertrophic scar fibroblasts(HSFB) were cultured in vitro,and fibroblast populated collagen lattice(FPCL) was set up.The effect of Hirudin on HSFB model with exposure to 0U/ml,1U/ml,10U/ml or 50U/ml Hirudin for 24,48 or 72 hours in vitro were studied.We detected the cells' vitality and proliferation by MTT method,measured the gels' contractile index and assessed the expression ofⅠ、Ⅲprocollagen mRNA by reverse transcription PCR.Results:Hirudin could inhibit fibroblasts' proliferation and vitality,decrease the contractile index of FPCL and diminish the expression of Ⅰ、Ⅲ procollagen mRNA.Conclusions:Hirudin has a distinguished inhibitory action to proliferation,secretion and contraction of HSFB in vitro,which may be the mechanism of inhibiting hypertrophic scar.
