OBJECTIVE A new simple method based on cloud-point extraction(CPE)and HPLC was developed to determine aloe-emodin,emodin and chrysophanol in rat plasma.METHODS The non-ionic surfactant Triton X-114 was chosen as the extract solvent.A Kromasil C18(4.6 mm×150 mm,5.0μm)column was used for gradient elution separation at 40℃ with detection wave length at 254 nm.RESULTS The concentration-response relationship was linear over a concentration range of 0.117-15 mg·L-1,0.258-16.5 mg·L-1 and 0.126-16.2 mg·L-1for aloe-emodin,emodin and chrysophanol, respectively. All calibration curves showed good linearity(r20.9997)within the test ranges. The intra-and inter-day variations for 3 analytes were 2.85%、3.27%、4.84%and 5.38%、6.01%、5.12%,respectively.CONCLUSION As a simple, less polluting and time-consuming method,it was successfully applied for the evaluation of pharmacokinetic profiles of aloe-emodin,emodin and chrysophanol in rat after administrating the extracts of Rhubarb.
