Detection of Classical Swine Fever,Porcine Parvovirus Infection and Porcine Pseudorabies by Multiplex Polymerase Chain Reaction

A multiplex polymerase chain reaction was optimized to simultaneously detect classical swine fever virus(CSFV),porcine parvovirus virus(PPV) and porcine pseudorabies virus(PRV).Three sets of specific primers were designed according to the conservative sequence E2 of CSFV,VP2 of PPV,and gⅡ of PRV in the gene bank.It was showed that all samples,which contained CSFV,PPV and PRV,could be amplified by the multiplex PCR using these three sets primers,with optimized factor of reaction.Three specific bands of CSFV 288 bp,PPV 575 bp,and PRV 700 bp,as little as 14 5 ng/L of CSFV,27 1 pg/L of PPV,and 31 4 ng/L of PRV were detected using agarose gel electrophoresis in this multiplex PCR.
