Orforglipron (LY3502970), a novel, oral non‐peptide glucagon‐like peptide‐1 receptor agonist: A Phase 1b, multicentre, blinded, placebo‐controlled, randomized, multiple‐ascending‐dose study in people with type 2 diabetes

Dear Editor, We wish to clarify differences between figure 3 of the main manuscript (Body weight change from baseline at week 12) and table S3 (Summary of pharmacodynamic parameters at baseline and week 12)—specifically that the least squares mean (LSM) change from baseline body weight values shown in figure 3 are different than the implied weight change values indicated in table S3 across all orforglipron doses, when subtracting the LSM of body weight at baseline and week 12.1 We can clarify that the differences between the values shown in figure 3 and table S3 are because of the differences in the analysis model. The LSMs presented in figure 3 reflect the analysis from a linear mixed model with change from baseline as the response, while table S3 represents data from a model with actual values at week 12 as the response. For the change from baseline analysis, an unstructured covariance structure was used (figure 3), while for the actual weight values (table S3), there were model convergence issues when the same covariance structure was attempted and, therefore, a compound symmetry covariance structure was applied. With this subtle difference, the treatment differences are slightly different between the two analyses. The study pre-specified analysis for the treatment comparison is based upon the change from baseline (figure 3), and thus it is recommended that readers use this for interpreting the treatment effects.
