Objective:To investigate the characteristics of immunophenotype of chronic lymphocytic leukemia(CLL). Method:Immunophenotyping was performed in 103 CLL patients by three color flow cytometry analysis using CD45 /SSC gating. Meanwhile 5 cases of T lymphocyte proliferative disease and 92 newly diagnosed acute lymphocytic leukemia (ALL) were selected in which there were only 5 cases of acute lymphocytic leukemia mature B (Mature-B-ALL) as control at the same time.Result:①In the 103 cases,almost all the CLL patients expressed CD19 and CD20,and the positive rate was 99.0% and 96.0% respectively. The positive rate of T-associate antigen CD2,CD3,CD4,CD7,CD8,CD25 and CD5 was 3.0%,3.0%,2.0%,2.9%,1.0%,18.6% and 78.6%,respectively. In 5 cases of T lymphocyte proliferative leukemia,the positive rate of both CD2 and CD5 were 80.0%,and none of B-associated antigens expressed. All the 108 patients expressed HLA-DR and none of the patients expressed CD1a,CD14,CD34 and CD57. ②CLL and T lymphocyte proliferative leukemia both had high expression of myeloid antigen. The positive rate of CD13 and CD11b were 51.1% and 42.9% respectively in CLL. While in T lymphocyte proliferative leukemia,the positive rate is 100% and 66.7% respectively. ③Compared with ALL-Mature-B,CLL had higher expression of CD5 (P0.05) but lower expression of cCD79a,CD10 and CD34(P0.05). There were no significant differences in the expression of HLA-DR,CD19 and CD20 among them. Conclusion:The sensitivity of CD19 and CD20 in diagnosis CLL is high,but low in specificity. The co-expressing of CD5/CD19/CD20 is the typical immunophenoty of CLL. CD5,CD10 and cCD79a can be used as the specific marker to distinguish B-CLL and Mature-B-ALL.
