The comparison study of Propylene glycol alginate sodium sulfate in combination with aspirin or clopidogrel for anticoagulant effect, antiplatelet effect and on the properties of blood rheology

Objective To compare the effect of Propylene glycol alginate sodium sulfate(PSS) in combination with aspirin or clopidogrel for anticoagulant effect,antiplatelet effect and on the properties of blood rheology.Methods 120 Wistar male rats were randomly divided into control group,aspirin group,clopidogrel group,PSS group,aspirin+PSS group,clopidogrel+PSS group.After 14 days' drug administration,blood was collected from abdominal artery for TEG(thromboelastography) test.At the same time,platelet-aggregation rate,platelet-adherence rate,erythrocyte sedimentation,hematokrit,and blood viscosity were tested.Results The value of R and K increased,while α-Angle,MA,and CI decreased in PSS group compared with control group.For aspirin+PSS group and clopidogrel+PSS group compared with aspirin group and clopidogrel group respectively,R and K increased and α-Angle,MA,CI decreased(F=25.95~82.83,P=0.000~0.012).The platelet-aggregation rate decreased in aspirin or clopidogrel plus PSS group compared with control group(χ2=25.207,v=5,P=0.000).Besides the decrease of erythrocyte sedimentation and blood viscosity(F=10.72,6.77;P=0.000,0.011),the platelet count reduced in PSS group compared with control group(F=6.98,P=0.009) but platelet-adherence rate and hematokrit had no difference(F=2.087,1.26;P=0.083,0.294).Conclusion PSS inhibited the coagulation factor and clot by fibrin and platelet.Moreover,aspirin or clopidogrel plus PSS can decrease CI and platelet-aggregation rate significantly.At the same time the thrombocytopenia induced by PSS didn't appear.In conclusion,the therapy by aspirin plus PSS,clopidogrel plus PSS may be better choices for anti-coagulant and antiplatelet treatment than aspirin or clopidogrel alone.
