Casparian strips were isolated from primary roots and needles by enzymatic isolation, the cellular structure was observed under the fluorescence microscope and chemical characterization was analyzed by Fourier transform infrared (FTIR) spectroscopy. The results indicate the presence of the primary pit fields in the longitudinal walls of net isolated from the needles, while such structure is not evident in the cell walls of the primary roots. The net from the needles appears to be more orderly, similarly sized, and well aligned;while those from the primary roots are irregular in sizes as well as mesh positioning. The FTIR analysis indicate that the Casparian strips from primary roots contain notably higher amount of suberin and lignin thanthose from the needles, whereas the content of cellulose from primary roots is lower than those from the needles. However,in terms of protein contents in the cell walls, one bears no significant difference from the other. The comparative study provides new reference for the study of physiological functions of Casparian strips in the endodermis of the leaves and roots.
