Polygonatum cyrtonema has been traditionally used as medicine and food in China for centuries, Polygonatum cyrtonema polysaccharides (PCP) is a principal constituent of P. cyrtonema and has been reported to have potential beneficial effects in alleviating high-fat diet (HFD)-induced obesity. The present study sought to identify the biomarkers and metabolic pathways associated with the anti-obesity effect of PCP through metabolomics. The partial least orthogonal squares-discriminant analysis (OPLS-DA) revealed significant differences in the metabolic profiles between the normal diet (ND) and HFD groups and HFD and PCP groups. A total of thirty biomarkers contributing to the effects of PCP on obesity were screened and identified. The analysis of metabolic pathways indicated that PCP prevented obesity by regulating linolenic acid, arachidonic acid, and glycerol phospholipid metabolism. The above results revealed the metabolic mechanism of PCP on obesity alleviation. Overall, this study establishes a novel approach of diagnostic and prognostic indicators for other diseases.
