OBJECTIVE To research whether CYP3A5 genetic polymorphism in kidney-transplanted patients were associated with blood concentration/administration dose of tacrolimus.METHODS 27 kidney-transplanted patients treated with tacrolimus were enrolled in this study.To establish ploymerase chian rection and allele-specific amplification(ASA) for study the distribution of CYP3A genetic polymorphism in 27 kidney-transplanted patients,and the blood concentration of tacrolimus was tested by enzyme linked immunosorbent assay(ELISA).RESULTS The oral tacrolimus dosage had great variation between individuals.The occurrence of the CYP3A5 genetic polymorphisms(A6986G) designated as CYP3A5*3 was fifty percent,The concentration/dose ratios of CYP3A5*1/*1 genotype and *1/*3 genotype patients were significantly 1ower than those of *3/*3 genotype patients(P0.01).There were mild differences between *1/*1 homozygotes and *1/*3 heterozygotes,but not statistically significant.CONCLUSION There's some relationship between CYP3A5 genetic polymorphisms and blood concentration of tacrolimus in kidney transplanted patients.If the same blood concentration of tacrolimus is presented in three genotypes,type of CYP3A5*1/*3 and CYP3A5*1/*1 should take higher dosage when it's compared to CYP3A5*3/*3 genotype.Analyzing the genetic polymorphisms of kidney-transplanted patients is good for designing individual tacrolimus dosage regimen in transplanted patients.
