Rigorous synthetic methods and potential biotoxicity limit the clinical transformation of metal-based nanomaterials (MNs). Herein, we proposed a novel one-step synthetic approach for silver nanowires (AgNWs) and improved their biosafety tremendously. Dopamine (DA) functioned as a reductant for AgNW formulation in the alkaline condition instantaneously and a protective agent by polymerizing to polydopamine (PDA) on the surface of AgNWs to form AgNWs@PDA. The protection of PDA on the AgNWs in situ enhanced the biosafety and photothermal conversion of AgNWs, which suggested potential for photothermal therapy in vivo. The results revealed that the AgNWs@PDA showed superior biosafety and antitumor efficacy than AgNWs. Overall, this work presents a new one-step synthetic approach for enhancing the biosafety of toxic MNs universally without weakening their surface plasmon resonance effect, which facilitates further clinical transformation.
