Aim Response surface methodology was employed to optimize prescription of the BSA freeze-dried liposome.Methods The effects and interaction of three factors on encapsulation efficiency and drug loading of freeze-dried liposome,such as cholesterol(CH)/phosphatidyl choline(PC),bovine serum albumin(BSA) concentration,trehalose concentration,were investigated by the method of central composite design.Quadratic regression models were set up according to the experimental data.And response surfaces were delineated on the basis of best-fit mathematic models.Results The optimal technology conditions of BSA freeze-dried liposome are as follows: the ratio of CH/PC is 0.25,BSA concentration is 3.40 mg·mL-1 and trehalose concentration is 33.00 mg·mL-1.Conclusion The results show that the encapsulation efficiency and drug loading of the BSA freeze-dried liposome are 63.10% and 26.91% respectively under the optimal conditions.And there are small deviations between the measured values of index and model predicted values.It shows that the response surface analytical method is reliable and can be used in the process of prescription optimization.
