The aims of this experiment were to optimize the extraction technology and to study the antioxidant activity of Castanea mollissima Blume polyphenols. The orthogonal test was adopted and the factors influencing the process parameters were determined. The antioxidant activity was evaluated through detecting DPPH· and ABTS+· scavenging rates, and peroxide value(POV) of Castanea mollissima Blume polyphenols in vitro. The results showed as follows: 1) optimal conditions for the extraction: ethanol concentration was 30%,liquid-solid ratio was 18∶ 1(mL∶ g),extraction temperature was 80 ℃,and extraction time was 140 min.The corresponding extraction yield was 22.61%,and the polyphenol content was 51.23%.2) The DPPH· and ABTS+· scavenging rates of polyphenols from Castanea mollissima Blume were similar to that of vitamin C,and higher than that of vitamin E.Castanea mollissima Blume polyphenols displayed an excellent antioxidant activity in the lard oil and the effect depended on its concentration, with the best concentration of 100 μg/mg.The antioxidant effect of Castanea mollissima Blume polyphenols at this concentration was equal to that of vitamin C,and both of them were better than vitamin E.In conclusion, under the optimal conditions,Castanea mollissima Blume polyphenols shows higher extraction yield and content and reveals better antioxidant activity.
