摘要
The alleviating effects of He-Ne laser on damage of nuclear actin filaments organization caused by ultraviolet-B (UV-B) light were investigated. Wheat (Triticum aestivum L. cv. ML7113) seedlings were irradiated with enhanced UV-B (10.08 kJ/(m2 d)), or He-Ne laser (5.43 mW/mm2), or combination of He-Ne laser and UV-B. The existence, distribution and relative content of F-actin in the nucleus were demonstrated by confocal microscopy and flow cytometry after fluorescence labeling with FITC-phalloidin. It was shown that the enhanced UV-B radiation significantly increased the diffuse fluorescence staining that resulted in bright fragments of F-actin being observed in the nucleoplasm. The damage caused by UV-B radiation was partially alleviated when the He-Ne laser was used: it could be seen that F-actin distribution in experiment was similar to that in the control.
