Significance of Expression of Cyclic Adenosine Monophosphate Response-Element Binding Protein in Children with Acute Lymphoblastic Leukemia

Objective To explore the role of cyclic adenosine monophosphate response-element binding protein(CREB) in children with acute lymphoblastic leukemia(ALL).Methods 1.Quantitative real-time reverse transcriptase polymerase chain reaction(RT-PCR) was used to examine the expression of CREB mRNA in the bone marrow of 10 children with not leukemia(NL)(including infection diseases or ITP),9 children were examined twice in different periods.2.The expression of total CREB and phosphorylated CREB in the bone marrow of children with ALL were detected by enzyme-linked immunosorbent assay(ELISA).Results 1.The expression of CREB mRNA in children with HR-ALL were significantly higher than that in children with NL or LR-ALL or MR-ALL(Pa0.05).They were 1.13±0.81,1.47±1.02,1.24±0.60,8.49±12.24,respectively.But the differences were not significant in the 3 latters.2.The expression of total CREB protein(ng/106cell)in HR-ALL children were significantly higher than that in LR-ALL or MR-ALL children(Pa0.05).They were 4.40±4.60,0.99±0.43,respectively.The expression of phosphorylated CREB(U/106cell)in HR-ALL children was significantly higher than that in LR-ALL or MR-ALL children(Pa0.05).They were 1.95±1.82,8.67±8.19,respectively.3.Sixty percent HR-ALL children had not less than one high risk factors according to MICM.The rate of overexpression of CREB was 55.0% in HR-ALL children.If combined the 2 sides,the rate of correspondence could be elevated to 85.0%.4.The rise of CREB mRNA level clued the disease did not release or relapsed.Conclusions The expression of CREB in children with HR-ALL is higher.The expression of CREB may be helpful for improving ALL phenotype and curative effect judgement.
