PROTOCOL 02-02.1: Storage on PDA Mineral Oil Slants

To store Pythium cultures using this method, transfer a pure culture onto PDA slants, grow for 3–5 days, and cover with sterile mineral oil. Store between 15 and 23 degrees C in the dark. For recovery, remove a piece of agar, blot off the oil, and plate on water agar. While most species remain stable, some may lose sporulation ability over long-term storage.
