Objective: To examine the effect of different culturing conditions on the development of mouse blastocysts in vitro. Methods:The mouse blastocysts on day 4 of pregnancy were respectively cultured in the serum-free medium(group Ⅰ),serum-need medium(group Ⅱ) or with mouse endometrium epithelial cells in serum-free medium(group Ⅲ).Then the rates of expanded blastocyst and hatching blastocyst in the different groups was compared. Results:Almost no blastocyst was seen to expand or hatch in groupⅠ,while the culturing systems of group Ⅱ and Ⅲ could surpport the development of embryonic cells. After 24 hours of culturing, the rates of expanded blastocyst and hatching blastocyst in group Ⅲ were much higher than that in group Ⅱ(P0.01).And the behavior of blastocysts similar to implantation was observed in group Ⅲ.Conclusion:The hatching and the development of blastocyst depends on the culturing conditions, while the co-culturing system of blastocysts with endometrium epithelial cells is much similar to the conditions in vivo, and can effectively increase the development of blastocysts.
