Therapeutic Effects of In Vivo Mobilizing and Inducing Bone Marrow Stem Cells on Diabetes of Rats

Objective To explore the possibility of mobilized bone marrow stem cells(BMSCs) induced by epidermal growth factor(EGF)and hepatocyte growth-promoting factor(pHGF) in vivo differentiating into insulin-producing cells and to test the therapeutic potency of insulin-producing cells(IPCs) on diabetes of rats.Methods 40 SD rats were randomly divided into four groups which were normal control group,untreated diabetes group,mobilization group and induction group with 10 rats in each group.Rats in the latter three groups received intraperitoneal injection of streptozotocin(STZ) to establish diabetes model.Rats in mobilization group and induction group were treated with subcutaneously injection of recombination human granulocyte colony stimulating factor(rhG-CSF,10 μg·kg-1·d-1) once a day for 15 days.Rats in induction group were treated with intraperitoneal injection of EGF(1 μg·kg-1·d-1) and pHGF(0.3 mg·kg-1·d-1) once a day for 14 days the day after mobilization injection.Before and after experiment blood glucose and serum insulin were measured,and insulin-positive cells in pancreas were detected by immunohistochemistry after experiment.Results Blood glucose level of induction group was significantly lower than that of the untreated diabetes group and mobilization group(P 0.05) while serum insulin was significantly higher(P 0.05).There were significantly more pancreatic insulin-positive cells in induction group than the untreated diabetes group and mobilization group(P 0.01).Conclusion Combination of EGF with pHGF after mobilization could induce bone marrow stem cells differentiating into insulin-secreting cells in streptozotocin-induced diabetic rats and improve hyperglycemia.
