Objective: To study the inhibition effects of silicone molecule on fibroblasts and the mechanisms of silicone gel sheet on hypertrophic scar. Methods: The scar-derived fibroblasts were incubated in culture medium with silicome oil of different concentrations (1 ∶ 40-1 ∶ 25 600) mixed and emulsified with Towen-20 by ultrasonic wave.The effects of inhibition of silicone molecule to fibroblasts were evaluate.Cytoactivity was determined by MTT. 102 healed burned-wounds of 51 patients were studied, 51 of 102 healed wounds were treated by silicone gel sheets to compare with the control groups. Results: ①The cytoactivity of fibroblasts was suppressed significantly compared with the PBS groups with different concentration of silicone oil ranged from 1 ∶ 3 200 to 1 ∶ 12 800 (P0.001). ②Excellent appearances and functions of healed wounds were examined for patients treatmented with silicone gel sheets at least 3 months compared with the control groups. Conclusion:Silicone gel sheets can help hypertrophic scar structure to be conversed to be structure of normal skin and improve the appearance and functions of the wounds healed.Silicone molecule may demonstrate the inhibition mechanism to the fibroblasts.
