Determination of Content of Rebaudioside A and Stevioside in Stevia rebaudiana Bertoni by RP-HPLC

HPLC method for the determination of rebaudioside A and stevioside in Stevia rebaudiana Bertoni is established by ultrasonic assisted extraction method.The separation is achieved on a Kromasil C18column(4.6mm×250mm,5μm)at 30 ℃ using methanol and water solution(75∶25,V/V)as the mobile phase at a flow rate of 1.0mL/min,210nm as the detection wavelength.Rebaudioside A shows good relationship at the range of 0.868~8.68μg(R=0.999),the recovery is 97.5%and RSD is 1.8%(n=6).Stevioside shows a good relationship at the range of 1.004~10.04μg(R=0.999),the recovery is 95.6%and RSD is 1.6%(n=6).The proposed method is accurate,simple and applicable for the determination of rebaudioside A and stevioside in Stevia rebaudiana Berton.
