Objective To observe the effect of oxymatrine(OM) on cell proliferation and COX-2 expression in human gastric cancer cell line BGC-823.Methods Human gastric cancer BGC-823 cells was cultured in vitro with oxymatrine, the MTT assay was used to determine cell viability at 24h,48 h and 72h.Western Blot and Real-time RT-PCR were applied to examine the protein and mRNA expression of COX-2.Results Low-dose(1.0 mg/mL) oxymatrine had no inhibitory effect on cell viability of BGC-823 cells.Oxymatrine significantly inhibited cell viability in a time and concentration-dependent manner when concentration exceeded 1mg/ml(P0.05),and down-regulated the protein and mRNA expression of COX-2(P0.05).Conclusion Within a certain drug concentration, oxymatrine can inhibit cell viability of BGC-823 cells and down-regulating the expression of COX-2.
