The processing of pressurized water extraction for extracting polysaccharide from Auricularia auricular was studied and optimized. The monose composition of the polysaccharide from fruiting body and mycelium of Auricularia auricular was determined by filter paper chromatography and compared after hydrolyzed the polysaccharide. The results showed that compared with traditional water extraction, the polysaccharide extraction ratio by pressurized water extraction was higher, and the most suitable conditions for extracting the polysaccharide from fruiting body were:90min extracting time,1∶35 solid-liquid ratio and 115℃ extracting temperature. Under these conditions the polysaccharide extraction ratio was 7.1g/100g. A single homogeneous material of the polysaccharide from fruiting body was glucose, and the extracellular polysaccharide from cultured mycelium mainly consisted of mannose.
