Abstract Recombinase polymerase amplification (RPA) is an isothermal amplification technology developed in recent years. The RPA technology not only offer fast detection speed, highly sensitivity and specificity, it can also amplify at room temperature. It is especially suitable for in the field and bedside diagnosis outside the laboratory and has a broad application. Here, we summarizes the development of RPA technology, describes the principle of RPA, reaction mechanism. reaction system and optimization of reaction design of RPA technology. Field diagnosis methods have developed on this basis, such as lateral flow dipstick, flocculation analysis, real-time fluorescence analyses, electrochemistry, chemiluminescence and surface enhanced Raman scattering. We also evaluate the "hot topics" in current development of RPA technology. Our review will provide references materials for further research and application of RPA technology. |
