Objective To prepare mouse monoclonal antibodies (mAbs) to dengue type 2 virus (TR1751) and identify their characte- ristics. Methods Suckling mouse brain homogenate containing high titer of dengue type 2 virus was centrifugated for removing cellular fragment and then used as antigen to immunize Blab/c mice. The spleen cells of immunized mice were fused with SP2/0 cells. ELISA, indirect immu- nofluorescent staining, plaque reduction neutralization test (PRNT), and Western blotting were employed for selection and identification of the antibodies. Results Three hybridoma lines, named I_1, I_9, and B_ 152 , were obtained, which could secrete mAbs. All of the mAbs were neutralizing antibodies and could recognized E protein of the virus. Conclusion The dengue virus special mAbs are prepared successfully, which is helpful for further studying the pathogenesis of this virus and developing the dengue virus vaccines as the immunological tools.
