Development change of ICAM-1 expression and protective effect of N-acetylcysteine in ischemia-reperfusion injuried rat retina

Objective:To study the development change of intercellular adhesion molecule-1 (ICAM-1)expression and the protective effect of N-acetylcysteine in ischemia-reperfusion injuried rat′s retina.Method:The model of retina ischemia-reperfusion was employed in 60 SD rats,which were randomly devided into two groups,one was ischemia-reperfusion group,the other was ischemia-reperfusion and N-acetylcysteine group,and each group was devided into 1 hour,6 hours,12 hours,24 hours,48 hours,72hours groups,with 5 rats in each group.The ICAM-1 was measured with in situ hybridization(ISH)method and immunohistochemial method in rat’s retina and ERG of each rat was examinated before been executed.Results:In ISH method,ICAM-1 mRNA began to express at 6 hours and expressed most at 24 hours after ischemia-reperfusion injury.The protein of ICAM-1 began to express at 12 hours after ischemia-reperfusion injury.In ischemia-reperfusion and N-acetylcysteine group,the ICAM-1 mRNA also began to express at 6 hours and the protein of ICAM-1 began to express at 12 hours after ischemia-reperfusion injury,but its level was lower than that of ischemia-reperfusion group at the same stage(P0.05).The b wave of ERG in ischemia-reperfusion group was lower than those in ischemia-reperfusion and N-acetylcysteine group at each stage (P0.05).Conclusions:ICAM-1 takes part in rat retina ischemia-reperfusion injury,and the N-acetylcysteine can suppress the expression of ICAM-1 relieving the retina from the ischemia-reperfusion injury.
