Previous studies have reported a tight interaction and cross-talk between ionotropic receptors, namely between P2XRs and other ionotropic receptors such as nAChRs, 5-HT3Rs and GABAARs. These interactions are all characterised by a physical interaction that leads to an activity-dependent cross-inhibition designed to negatively control the excess of function of other fast signalling systems by ATP. Since P2X and NMDA receptors are coincidence detectors of neuronal function, a putative dimerization between P2X and NMDA receptors would be of particular interest. This hypothesis is prompted by the observations that ATP and glutamate are co-released from nerve terminals and P2XRs and NMDARs have an overlapping subcellular localization in the CNS.We now report the existence of hybrid P2X2/NR2B receptors, ionotropic receptors composed by subunits from dissimilar receptor families that are able to sense more than one signalling molecule.
