Objective To investigate the influences of three different hemi-extracorpreal liver resection on the liver ischemia-reperfusion injury. Methods Forty Sprague-Dawley rats were divided into 4 groups equally (A,B,C and D).Blood and liver specimens were collected at 4th h and 24th h after liver reperfusion, respectively.Specimens were subjected to HE staining, test of the serum concentration of ALT,RT-PCR,and immunohistochemistry assay. Results Liver function test showed that ALT was increased after reperfusion and its extent was depended on the complexity of the operation and the reperfusion time.In HE staining, the extent of inflammatory reaction was also related to the complexity of the operation and the reperfusion time.In group B,there was no difference between the expression of PPARα at 4th h postoperative liver tissue and 24th h postoperative liver tissue, but the expression of bcl-xL was increased significantly after 24 h reperfusion. In groups C and D,the expression of bcl-xL and PPARα was increased after 4 h reperfusion, but 24 h after reperfusion, the expression of PPARα was increased significantly as compared with that at 4th h postoperative liver tissue, and the expression of bcl-xL was lower than that at 4th h postoperative liver tissue. Conclusion PPARα and bcl-xL play very important roles in the liver reperfusion injury after the hemi-extracorpreal liver resection and their expression reflects the extent of the liver reperfusion injury.
