Comparative Study on Collagen Fiber Staining between Sirius Red in Saturated Carbazotic Acid Staining and MASSON Trichrome Staining on Rat Kidney Specimen

Objective To compare the effect of collagen fiber staining between Sirius Red F 3B(SR) in saturated carbazotic acid staining and MASSON trichrome staining on rat kidney specimen.Methods To establish renal interstitial fibrosis model of half ureteral obstruction in rat,then to get kidney specimen in obstruction side and carry out Sirius Red F 3B(SR) in saturated carbazotic acid staining and MASSON trichrome staining.Eventually,we measured the area of renal interstitial collagen by imaging analysis software.Results Both Sirius Red F 3B(SR) in saturated carbazotic acid staining and MASSON trichrome staining displayed collagen fiber.They had no significant difference in measuring the area of renal interstitial collagen.Furthermore,Sirius Red F 3B(SR) in saturated carbazotic acid staining could distinguish the collagen types through polarizing microscope.Conclusion The operation procedures of Sirius Red F 3B(SR) in saturated carbazotic acid staining are simpler than those of MASSON trichrome staining and it is a fine method to display collagen.
