Objective To establish a hyperglycemic clamp technique,a method for the precise measurement of the insulin secretion of β-cell . Methods A hyperglycemic clamp technique was used to the study of methodology in 12 normal subjects.Results (1)The plasma glucose concentration of the individuals reached a desired hyperglycemic within the fixed time(14min) and maintained the hyperglycemic plateau (13.05±0.18)mmol/L during the whole clamp;(2)Under these conditions of constant hyperglycemia, the insulin response is biphasic with phase 1 of (295.38±20.92)mU/L,and phase 2 of (83.13±5.18)mU/L; (3) The insulin sensitivity index (13.23±1.98)mg·kg -1 ·min -1 /mU/L form the hyperglycemic clamp was higher than that from the euglycemic clamp. Conclusion (1) The hyperglycemic clamp technique was established successfully; (2) The hyperglycemic clamp allows for obtaining value of the two-phases of insulin release and maximum insulin secretion to evaluate β-cell function;(3)The hyperglycemic clamp can also measure insulin sensitivity to assess insulin resistance.
