The influence on the function of peritoneal macrophage in mouse with cyclophosphamide

Objective To study the influence on the acid phosphatase of peritoneal macrophage with cyclophosphamide.Methods Twenty mice were divided into experimental group and control group randomly,ten mice in each group.Cyclophosphamide(50 mg·kg-1)and physiological saline(0.5 mL)was injected into the back with subcutaneous injections in the experimental group and in the control group respectively.The two groups all were one time per day for 5 days continuously.The mice in two groups were immunized with starch broth in the third day of the injection with one time per day.After the third immunization for 3~4 h,the peritoneal fluid were taken out and incubated for 30 min in acid phosphatase action liquid at 37 ℃.Then they were fixed with formalin and acted with ammonium sulfide,rinsed with distilled water,open-air drying then they were examined with microscope.Results After hypodermic with cyclophosphamide,the activity of acid phosphatase in peritoneal macrophage in experimental group was decline obviously.Average density image analysis of macrophage indicated that the average density of control group was 127.36±9.52 and experiment group was 65.37±6.37.And there was significant deviation between the two groups(P0.01).Conclusion Cyclophosphamide has the inhibitation to the nonspecific immunity system of mouse.
