Purification, Crystallization and Amino Acid Composition of α-l-Arabinofuranosidase from Aspergillus Niger

Crystalline α-l-arabinofuranosidase was obtained from the culture fluid of Aspergillus niger Kz. An improved method was used consisting of heat treatment, chromatography on DEAE-cellulose, DEAF-Sephadex A-50 and SE-Sephadex C-50, and gel-filtration on Sephadex G-100. The molecular weight of the purified enzyme was found to be about 53 000 by the procedure of gel-filtration. The isoelectric point lies near pH 3.6. The amino acid composition is shown in Table 11.
