Разработка модели противоопухолевой липосомальной вакцины

The aim of this study was to develop model anticancer liposomal vaccine containing a lysate of rat glioma C6. We have optimized the method of cell lysate preparation to get a small single-layer liposomes. The size of liposomes containing a lysate of rat glioma C6 was 185±10 nm, the percent of the included protein was 60±5%. We have confirmed by gel-filtration that the efficacy of cell lysis was increased from 5-10% to 90-95% if we lysed cells using ultrasound technique in addition to freezingthawing method of cell lysis. We have also shown by Western Blot analysis that ultracentrifugation of cell lysate does not influence the level of membrane antigenes, tested using other approach for cell lysis.
