Effect of Shengfu Injection on Peripheral T-lymphocyte Subsets and Natural Killer Cell in Patients with Severe Sepsis

Objective:To investigate the effect and clinical significance of Shengfu Injection on peripheral T-lymphocyte subpopulations and natural killer cell(NK cell) in patients with severe sepsis.Methods:Sixty-eight patients with severe sepsis were divided into a control group(n=32) and a treatment group(n=36).The patients of two groups were all treated conventionally,and the only difference was that Shengfu Injection was given to the treatment group at the early stage.The T-lymphocyte subsets and NK cell counts(CD+16+56) in the peripheral blood were detected respectively before and 3 d,7 d after treatment by flow cytometry.10 cases were chosen as the health control group at the same phase.Results:There were insignificant differences of the T-lymphocyte subsets and NK cell counts between Shenfu group and control group before treatment(P0.05).Compared with the health control group,the T-lymphocyte subsets and NK cell counts were lower significantly in the two treatment groups.And they were raised in different extent between the two groups at the 3rd and 7th day after therapy.The CD3+,CD4+,CD8+T-lymphocytes and NK cell of peripheral blood in Shenfu group were all markedly elevated at 7th day after treatment(CD3+:498±248 vs 663±291,CD4+:302±197 vs 378±284,CD8+:208±103 vs 278±120,NK:170±75 vs 275±50,all P0.05).Furthermore,compared with the control group,the peripheral CD3+,CD4+,CD8+T-lymphocytes and NK cell of Shenfu group were elevated obviously at the 7th day after therapy,the differences were statistically significant(CD3+:663±291 vs 579±302,CD4+:378±284 vs 320±233,CD8+:278±120 vs 230±138,NK:275±50 vs 256±42,all P0.05).The 28-day mortality in Shenfu group and control group was 25% and 37.5%,respectively,and no significant difference between the two groups was found(P0.05).However,there was significant difference of intensive care unit(ICU) stay time [(10.5±3.2)d vs(12.2±2.8) d]between the two groups(P=0.012).Conclusion:In the treatment of patients with severe sepsis,on the basis of conventional therapy,the early use of Shengfu Injection can significantly improve the inhibition of T-lymphocyte subsets and NK cell,and help to maintain a balanced profile of immunologic function,and shorten the length of stay in ICU.
